Amifostine (WR2721) is a potent cytoprotective and radioprotective agent, selectively safeguarding normal tissues from chemotherapy and radiation-induced damage. It acts as a prodrug, activating to WR-1065 by alkaline phosphatase, which scavenges free radicals and protects cells. Amifostine also induces HIF-α1 and p53, key regulators in the cellular response to hypoxia and DNA damage. Its antiangiogenic properties, by modulating VEGF-A, suggest potential in inhibiting tumor vasculature.
